One breast significantly and uncomfortably bigger than the other.

Some women do have significantly assymetrical breasts. You haven't said how old you are, nor whether your breasts have always differed in size.

If this is not a recent or sudden change then it is very unlikely to signal underlying disease. If there has been a sudden increase in size, or if you have felt a lump then you should see a doctor about it.

If the difference between the breast is very noticeable, surgery could be an option.
